This product is a neutralizing human CD47 antibody derived from hybridomas. antibody used in various therapeutic approaches, CD47 is a ubiquitously expressed transmembrane glycoprotein with a single Ig-like domain and five transmembrane regions that serves as a cellular ligand for SIRPa through the NH2-terminal V-like structure of SIRPa Domain mediates binding.

Figure 1 Anti-Human CD47 Antibody in WB
Western blot analysis of HPAB-0008-FY was performed by loading Recombinant Human CD47 Protein (lane 1, non-reducing antigen: 1 μg) onto a 12% Tris-HCl polyacrylamide gel. Proteins were transferred to a CN membrane and blocked with 5% skim milk for at least one hour. Membranes were probed with HPAB-0008-FY and Goat Anti-Human IgG-HRP as a secondary antibody. Chemiluminescent detection was performed.

ELISA analysis of HPAB-0008-FY was performed by coating with Recombinant Human CD47 Protein. Then blocked with BSA, and incubated with Anti-Human CD47 Antibody (HPAB-0008-FY). The Goat Anti-Human IgG-HRP as a secondary antibody. Detection was performed using TMB substrate and stopped with sulfuric acid. The absorbances were read on a spectrophotometer at 450 nm.

SDS-PAGE analysis of HPAB-0008-FY in non-reduced (Lane 1) and reduced (Lane 2) conditions. Gel stained for 30 minutes with Coomassie Blue. As a result of different β-mercaptoethanol-reduced proteins (Heavy chain and Light chain) migrate as about 50 kDa and 25 kDa, respectively.
